Study: Genetic and functional analysis of archaeal ATP-dependent RNA ligase in ribosomal RNA biogenesis

In this study, we generated allelic knockouts of ATP-dependent RNA ligase (Rnl) in hyperthermophilic archaeon T. kodakarensis and analyze the total and small RNA by RNA-Seq. Overall design: Knocked out the TK1545 gene by homologous recombination, and prepared RNA samples for whole transcriptosome and small RNA seq analysis. The differentially expressed genes between the two strains were analyzed.
